OXTR is the receptor for oxytocin, a hormone tied to bonding and social behavior. A much-discussed variant has been weakly linked to empathy, but the evidence is shaky, so treat it as fun rather than fact.

What this variant is, and what it became

OXTR is the receptor for oxytocin, a hormone involved in birth, breastfeeding, social bonding and stress responses. This variant sits in an intron rather than in the protein-coding sequence, so it does not change the receptor itself, and no mechanism connecting it to behaviour has been established.

It nonetheless became one of the most studied variants in social neuroscience, appearing in hundreds of papers on empathy, parenting, trust, loneliness and stress, and in popular writing as the empathy gene.

What the evidence actually shows

Early studies reported that people with two copies of the G allele scored higher on empathy measures, and the finding spread quickly. Replication has been mixed ever since. One meta-analysis combining 13 samples and 6,631 people did find an association with some empathy measures. A later systematic review of 20 cohorts and 8,933 people found an effect only in young to middle-aged adults, and no significant overall effects across empathy subdomains.

This site's evidence file records no genome-wide association study for this variant. That absence is meaningful: traits with real common-variant signals usually show up when hundreds of thousands of people are studied without a prior hypothesis, and this one does not.

The frequency problem

In 1000 Genomes reference data, about 46% of European-ancestry individuals and 61% of African-ancestry individuals carry two copies of the so-called empathy allele, against roughly 12% of East Asian-ancestry individuals.

Any account of this variant as a marker of empathic ability has to explain why the genotype is a clear majority in some populations and a small minority in others, and no such account exists. That mismatch alone is a reason to hold the trait claim loosely.

Why this page is more about method than about you

This variant is a good example of the candidate gene era in behavioural genetics, when researchers picked a plausible gene, tested it in a few hundred people and published the associations that reached significance. A 2019 analysis of the 18 candidate genes most studied for depression found no support for any of them in large samples, and the field has largely moved to genome-wide methods for this reason.

Read your result here as history of science rather than as a description of your character. Empathy is also measured directly, if imperfectly, by questionnaires and by how people behave, both of which carry more information than this genotype.

What each rs53576 genotype means

rs53576 has three possible genotypes: AA, AG and GG.

Slightly lower empathy (rs53576 AA). You have two A copies of the oxytocin-receptor gene OXTR. Some studies have tied this to slightly lower scores on empathy or stress-handling measures, but the research is shaky and inconsistent, so treat it as a fun footnote rather than a real readout of your personality.

Typical empathy (rs53576 AG). You carry one A and one G copy of the oxytocin-receptor gene. This sits between the versions some studies have linked to social behavior, but the evidence is weak overall, so don't read much into it.

Slightly higher empathy (rs53576 GG). You have two G copies of the oxytocin-receptor gene OXTR, which a few studies have linked to slightly higher empathy or sociability. The research is contested and the effect, if real, is tiny, so enjoy it as trivia rather than fact.

Common questions

Does this variant make me more or less empathetic? The evidence does not support reading it that way. Meta-analyses disagree, no genome-wide study supports the variant, and the genotype called empathic is a majority in some populations and a minority in others.

Why is it called the empathy gene then? Because early small studies found an association and the name stuck in popular coverage. It is a striking example of how a candidate-gene finding can outlive the evidence for it.
