COMT "warrior vs worrier" (stress & dopamine)

COMT clears dopamine from the thinking part of the brain. A common variant slows it down: the slower 'Met' version is nicknamed 'worrier' (a touch more stress-sensitive, sometimes sharper focus), the faster 'Val' version 'warrior'. The real effects are small and heavily hyped, so treat it as fun.
What COMT does
COMT breaks down dopamine and related transmitters, and it matters most in the prefrontal cortex, where there are few dopamine transporters to do the job instead. This variant changes valine to methionine at position 158, and the methionine version of the enzyme is less stable at body temperature, clearing dopamine roughly three to four times more slowly.
That is a genuine, measurable biochemical difference. What has not held up is the behavioural story built on top of it.
Where warrior and worrier came from
The popular framing holds that fast-clearing valine carriers cope better under stress, the warrior, while slow-clearing methionine carriers have better working memory in calm conditions but suffer more under pressure, the worrier. It came from a real hypothesis about an inverted-U relationship between prefrontal dopamine and cognitive performance.
The framing is memorable and the evidence behind it is weak. Individual studies have gone in both directions, effect sizes shrink as samples grow, and the labels have travelled far beyond anything the data supports.
What large studies found
COMT was among the 18 most-studied candidate genes examined in a 2019 analysis across multiple large samples, which found no support for the historical candidate gene hypotheses for depression, either as main effects or in combination with environmental stress. ClinVar classifies this variant as benign, and the site's evidence file records no genome-wide association study for it.
The variant is also extremely common. In 1000 Genomes reference data about 27% of European-ancestry individuals carry two methionine copies and 50% carry one, so any trait framing has to apply to a very large fraction of everyone.
What this variant is still good for
As biochemistry, it is real and useful: COMT genotype does shift prefrontal dopamine clearance, and it appears in pharmacological research, including work on drugs that inhibit COMT in Parkinson's disease.
As a description of personality or stress tolerance, treat it as a story. How you respond to pressure depends on sleep, training, experience, context and what is at stake, none of which a single enzyme variant captures.
What each rs4680 genotype means
rs4680 has three possible genotypes: GG, AG and AA.
Val/Val ('warrior') (rs4680 GG). You have two 'Val' copies of COMT, so you clear dopamine from your prefrontal cortex quickly. This is nicknamed the 'warrior' type, loosely tied to steadier stress handling (and, in some studies, slightly less sharp working memory at rest). The effect is small and the labels are pop-science, so enjoy it as trivia.
Balanced (Val/Met) (rs4680 AG). You carry one 'Val' and one 'Met' copy of COMT, an in-between dopamine-clearing speed. Most people sit somewhere around here, and this gene's effect on stress or focus is small either way.
Met/Met ('worrier') (rs4680 AA). You have two 'Met' copies of COMT, so you clear prefrontal dopamine slowly, the 'worrier' nickname, loosely linked to a bit more stress-sensitivity and, in some studies, sharper working memory. The science is contested and the effect is small, so take the label lightly.

Common questions
Am I a warrior or a worrier? Neither label is supported well enough to apply to a person. The enzyme difference is real, but the behavioural framing built on it has not replicated in large samples, and a 2019 analysis found no support for COMT in depression.
Does COMT affect how I handle stress? Not in a way that can be read off a genotype. Individual studies have found effects in both directions, and the variant is carried in some form by roughly three quarters of people of European ancestry.
