BDNF is a protein that helps brain cells grow and form memories. A common variant (Met66) slightly lowers its release during activity and has been weakly linked to memory and stress responses. The evidence is mixed, so read it lightly.

What the BDNF Val66Met variant does

BDNF makes brain-derived neurotrophic factor, a protein that supports the survival of neurons and the strengthening of synapses, and which is central to how memories form. This variant changes valine to methionine in the precursor protein, and laboratory work shows the methionine version is packaged and released from neurons less efficiently in response to activity.

That is a real cellular difference, and it is why the variant has been studied so heavily in memory, depression, anxiety and response to antidepressants.

Why the human findings are so much weaker than the cell biology

This variant was one of the historical candidate genes examined in a 2019 analysis across multiple large samples, which found no support for the candidate gene hypotheses for depression, including gene-by-environment versions of them. ClinVar classifies it as benign, and the site's evidence file records no genome-wide association study for it.

Imaging and memory studies have reported differences by genotype, often in small samples, and the effects shrink or vanish in larger ones. The gap between a well-demonstrated molecular effect and an unreliable behavioural one is the recurring theme of this part of the catalog.

A frequency difference that complicates every trait claim

In 1000 Genomes reference data, roughly 69% of East Asian-ancestry individuals carry at least one methionine copy, with 20% carrying two, against about 34% and 4% of European-ancestry individuals, and under 7% of African-ancestry individuals.

Any claim that this variant impairs memory or raises depression risk has to contend with the fact that most people in East Asia carry it. Trait claims that ignore population frequency usually collapse on contact with it.

What is real here

BDNF itself matters. Blood BDNF levels rise with aerobic exercise, and the protein is one of the better-supported mechanisms behind exercise being good for the brain. That is a fact about the protein, not about this variant, and it applies to everyone.

What each rs6265 genotype means

rs6265 has three possible genotypes: CC, CT and TT.

Val/Val (typical BDNF) (rs6265 CC). You have two 'Val' copies of BDNF, the common version, so your activity-driven release of this brain-growth protein is in the typical range. This gene's links to memory and mood are weak, so there's little to read into here.

One Met copy (BDNF) (rs6265 CT). You carry one 'Met' copy of BDNF, which slightly lowers how much of this brain-growth protein is released during activity. It's been weakly linked to memory and stress responses, but the evidence is mixed, so it's a soft signal at most.

Met/Met (BDNF) (rs6265 TT). You have two 'Met' copies of BDNF, tied to somewhat lower activity-dependent release of this brain-growth protein. Some studies link it to memory or stress differences, but findings are inconsistent, so treat it as a gentle hint, not a fact.

Common questions

Does Val66Met affect my memory? Not in any way you could rely on. Small studies have reported differences, larger analyses have not supported them, and most people of East Asian ancestry carry the methionine version.

Does this variant affect antidepressant response? It has been studied for that and the results have not been consistent enough to guide treatment. No pharmacogenomic guideline uses BDNF genotype.
