The MC1R gene shifts pigment toward red hair, fair skin and freckles. Red hair usually appears only when two MC1R variants combine, so several of these variants are counted together.
What MC1R does
MC1R is the switch on pigment cells that decides which kind of melanin they make. When it is working and stimulated, cells make eumelanin, the brown-black pigment that absorbs ultraviolet light. When it is impaired, they default to pheomelanin, the red-yellow pigment, which gives red hair and pale skin and provides much less ultraviolet protection.
The four variants read here are the common loss-of-function versions in European populations. Red hair typically appears when two are present, which is why the panel reads them together rather than one at a time.
Why it is more than a hair colour gene
MC1R variants raise melanoma risk, and they do so partly independently of hair colour: carriers with brown hair still show increased risk. Two mechanisms are involved, less ultraviolet shielding from pheomelanin and a pheomelanin pathway that appears to generate oxidative damage even without sun exposure.
That makes this one of the few appearance traits on the site with a genuine health footnote. The response is the ordinary one, sun protection and getting new or changing moles checked, and it applies more strongly to people who burn easily whatever their genotype says.
The anaesthetic question, in proportion
A widely repeated claim holds that redheads need more anaesthetic. It comes from a 2004 study that found women with red hair required about 19% more desflurane than dark-haired women, and related work reported greater thermal pain sensitivity and reduced response to subcutaneous lidocaine.
The finding is real but small in scale: that study compared ten redheads with ten controls. It is worth mentioning to an anaesthetist as a point of interest rather than treating as an established dosing rule, and MC1R is not much expressed in the central nervous system, so the mechanism remains unclear.
How common these variants are
Individually they are not rare in European-ancestry populations: the most common of the four is carried by roughly 17% of European-ancestry individuals in reference data, while all four are close to absent in East Asian and African-ancestry samples.
Because red hair usually needs two variants together, it stays uncommon even where the individual variants are frequent, and a single copy more often produces fair skin, freckling or a red tint in beard or body hair than red hair itself.
What this panel does not cover
MC1R is unusually variable, with dozens of known loss-of-function versions, and this panel reads four. Red hair is therefore still possible with a no-variant result here, particularly outside European-ancestry populations. Hair colour also depends on other genes, including the HERC2 and OCA2 region this site reads for eye colour, and on age, since red hair often darkens over time.
What each MC1R result means
Red hair likely. You carry two or more MC1R red-hair variants, which strongly favors red or strawberry-blond hair, fair skin and freckles. Fair, freckle-prone skin burns more easily, so sun protection helps.
One red-hair variant. You carry a single MC1R red-hair variant. Alone it usually means slightly fairer skin or a few freckles rather than red hair, which typically needs two such variants together.
No red-hair variant. You don't carry the MC1R red-hair variants this panel reads, so red hair is unlikely from MC1R. A few rarer MC1R variants aren't tested here.
