FOXO3 helps cells resist stress and is one of the very few genes linked, across many populations, to living to an exceptionally old age. The longevity-associated version slightly raises the odds of reaching very old age, though lifestyle and chance dominate.

What the FOXO3 longevity variant is

FOXO3 is a transcription factor at the end of the insulin and IGF-1 signalling pathway, the pathway that extends lifespan in worms, flies and mice when it is turned down. FOXO3 activation switches on stress resistance, DNA repair and autophagy programmes, which is why the gene has been a longevity candidate for decades.

This variant sits in an intron rather than in the coding sequence, so it marks a signal in the gene rather than changing the protein.

How strong the evidence actually is

The original finding came from a nested case-control study of Japanese-American men in Hawaii in 2008, and it is one of the very few longevity associations that has replicated at all. A Southern Italian centenarian study replicated it through a proxy variant with an odds ratio of 1.51, and a later analysis pooled four centenarian studies.

The effect is stronger in men than women across several of these studies. It is also worth noting what is absent: this site's own evidence file records no genome-wide association entry for this variant, which reflects how much smaller and more specialised the longevity literature is than the disease literature elsewhere on this site.

Why the frequency should temper any excitement

The allele associated with longevity is not rare or special. In 1000 Genomes reference data, about 38% of European-ancestry individuals and 46% of East Asian-ancestry individuals carry two copies, and roughly another 45% carry one.

A variant carried by most of humanity cannot be doing much to distinguish who reaches 100. Studies find it enriched in centenarians relative to controls, which is a statistical shift in an extreme group, not a personal forecast.

What this variant does not tell you

Longevity is the least tractable trait on this site. Twin studies put the heritable component of lifespan at roughly 20% to 30%, most of which is not attributable to any identified variant, and much of what looks heritable in family data reflects shared environment and the tendency of people to have children with people like themselves.

Nothing follows from this result. There is no intervention, no screening and no plan that changes with the genotype, and the things that do shift healthy lifespan, not smoking, physical activity, sleep, blood pressure and social connection, apply identically either way.

What each rs2802292 genotype means

rs2802292 has three possible genotypes: TT, GT and GG.

Typical (no FOXO3 longevity variant) (rs2802292 TT). You don't carry the FOXO3 longevity-associated version, so as far as this gene goes, your odds are at the typical baseline. Longevity is overwhelmingly about lifestyle, environment and chance, with genetics a minor player outside extreme old age.

One FOXO3 longevity copy (rs2802292 GT). You carry one copy of the FOXO3 version linked to reaching very old age. It's a small, favorable nudge tied to cellular stress resistance, and one of the few longevity genes replicated across populations. Habits still matter far more.

Two FOXO3 longevity copies (rs2802292 GG). You have two copies of the FOXO3 longevity-associated version, the genotype most enriched among people who reach 95 or 100. The effect on any one person is small and lifestyle dominates, but it's a genuinely interesting and well-replicated longevity signal.

Across global populations, about 28% of people carry two copies of this variant, and about 28% carry none.

Common questions

Does this variant mean I will live longer? No. It has been found more often in centenarians than in controls in several studies, particularly in men, but it is carried by most people, and lifespan is only modestly heritable to begin with. It is one of the least actionable results on this site.

Is FOXO3 the longevity gene? It is the most replicated longevity candidate, which is a much weaker claim than the nickname suggests. Aside from APOE, essentially no genetic variant has a large, reproducible effect on human lifespan.
